miRNA-Based Signature Associated With Tumor Mutational Burden in Colon Adenocarcinoma

Colon adenocarcinoma (COAD) is one of the most common malignant tumors. Tumor mutation burden (TMB) has become an independent biomarker for predicting response to immune checkpoint inhibitors (ICIs). miRNAs play important role in cancer-related regulation. However, relationship between miRNA expression and TMB COAD remains unclear. Therefore, transcriptome profiling data, clinical annotation profiles cases were downloaded from TCGA database. Subsequently, 323 randomly divided into training test sets. The differential high low groups set was obtained as a signature using least absolute shrinkage selection operator (LASSO) logistic regression verified set. Based on LASSO method, principal component analysis (PCA), ROC, we found that credible because it can discriminate levels. In addition, correlation 18-miRNA-based checkpoints performed, followed by qRT-PCR, measure relative 18 patients. miRNA-based model had strong positive with weak CTLA4 CD274 (PD-L1). no observed SNCA (PD-1). Finally, enrichment performed explore their biological functions. results demonstrated involved process immunity cancer pathways. conclusion, effectively predict different levels provide guide its treatment ICIs.
